Same issue here.
I would say it happens always when You try to overwrite the filename with exactely the same name but just change from upper case to lower case. Or vice versa.
Minimal change in filename (deletion of "space" for example) works pretty well.

just tested: That's perfectly it! Created a non-animated 2x2 preview (as jpg) and kept the existing animated custom-tiled (webm). Works just as described :-) - in multimedia preview is see the 2x2 - when I hover with the mouse, the animation is running - when open the screenlist with the overlay-but...

ok, let me rephrase: For me I don't see a difference whether the option is on or off.

- In the fullscren view of the screenlist I can capture a frame on the fly in both cases.

- And in the multimedia view I cannot capture a frame. There is no additional item in the RMB menu or whatever.
